Double Antibody Sandwich Method

1. Coating: The antibody is diluted to a concentration of 1–10 μg/ml using 0.05 M Phosphate Buffered Saline (PBS) at pH 9.6. Then, 0.1 ml of the solution is added to each well of a polystyrene plate and incubated overnight at 4°C. The next day, the liquid is removed, and the wells are washed three times with washing buffer for 3 minutes each time.

2. Sample Addition: Add 0.1 ml of the sample to the coated wells and incubate at 37°C for 1 hour. At the same time, include blank, negative, and positive control wells.

3. Enzyme-Labeled Antibody Addition: Add 0.1 ml of freshly prepared enzyme-labeled antibody (diluted according to titration) to each well. Incubate at 37°C for 0.5 to 1 hour, then wash the wells thoroughly.

4. Substrate Addition: Add 0.1 ml of TMB substrate solution to each well and incubate at 37°C for 10–30 minutes to develop color.

5. Stop Reaction: Add 0.05 ml of 2M sulfuric acid to each well to stop the reaction.

6. Result Interpretation: Results can be visually assessed on a white background. A darker color indicates a stronger positive result, while a negative result appears colorless or very light. The degree of color change can be marked as “+” or “-”. Alternatively, the OD value can be measured using an ELISA reader at 450 nm (or 410 nm if ABTS is used). If the OD value exceeds 2.1 times that of the negative control, it is considered positive.
